A few years ago, a team of British researchers decided to revisit the facts of King George III’s madness with a new analytic tool. They loaded some 500 letters written by the king into a machine-learning engine and laboriously trained the system to recognize various textual features: word repetition, sentence length, syntactical complexity, and the like. By the end of the training process, the system was able to predict whether a royal missive had been written during a period of mania or during a period of sanity.

So far, as you might have noticed, this sounds a lot like the Bayesian idea of the brain as an “inference engine” that Hinton told Friston about in the 1990s. And indeed, Friston regards the Bayesian model as a foundation of the free energy principle (“free energy” is even a rough synonym for “prediction error”). But the limitation of the Bayesian model, for Friston, is that it only accounts for the interaction between beliefs and perceptions; it has nothing to say about the body or action. Yes. I’d say, Steve, going forward, as we move even more of the revenue base on that, it definitely is a lower cost to serve. It’s a natural for us where we have over half our revenues with major customers that understand the efficiency of e-commerce. So we have multiyear MRO contracts with them. They have cross-referenced parts. We have pricing established. The terms and condition all established. So it really lends itself to the ease for them to order in their system, cross-reference to our catalog and places the order into our system very seamlessly. So it’s part of our model. We’ll deal with them, all of our customers at a transactional basis and the branches if that’s the preferred, we’ll do the multi-year contracts and large purchases for them. And we’ll also offer, with the items we have in stock for them and for their contract, really easy way for them to do business with us through the catalog. So I think it’s a trend that’s going to continue and we are investing to even enhance that better as I mentioned. So I think that is a growing part of our business mix.

HIV-1 Vpr can trigger G2 cell cycle arrest in the absence of either CUL4A or CUL4B but not both. (A) Cultures of HEK293T cells or HEK293T cells that stably express shRNA against firefly luciferase (control), CUL4A, or CUL4B were tested for CUL4-specific protein depletion by Western blotting. (B) HEK293T and HEK293T stably shRNA-expressing cell lines were mock infected or infected with HIV-1 or HIV-1 lacking Vpr at equivalent MOIs. Forty-eight hours after infection, cell nuclei were harvested, treated with RNase A and propidium iodide, and analyzed for DNA content by flow cytometry. (C) The ratio of nuclei with 4N DNA content (G2+M) to those with 2N DNA content (G1) was determined by using FlowJo software for replicates of the experiment shown in panel B. (D) HEK293T cells that stably express CUL4A-specific shRNA were depleted of CUL4B by transduction of a CUL4B-specific lentiviral expression vector. Transduction with a vector for CUL4A-specific shRNA was used as a control. Cultures under each experimental condition were tested for specific CUL4 protein depletion by Western blotting. (E) The HEK293T cultures described above for panel D were either mock infected or infected with HIV-1 or HIV-1 lacking Vpr. Nuclei from these cells were harvested 48 h after infection and analyzed for DNA content by flow cytometry, as described above for panel B. (H) HEK293T cells, as described above for panel G, were either mock infected or infected with HIV-1 or HIV-1 lacking Vpr.
